The Microbial test bottle is a sulfate reducing bacteria test tube that contains tubes of culture media specifically formulated to promote the growth of anaerobic sulfate reducing bacteria. It allows for the evaluation of both liquids and surfaces.

The most common sulfate reducing bacteria associated with contamination of the industrial environment are found in the genera Desulfovibrio and Desulfotomaculum. When sulfide is liberated, it reacts with iron in the tubed culture medium to form iron sulfide, a black precipitate. The degree to which the medium blackens, along with the length of time it takes to change color, allows for an estimated count of sulfate reducers to be made

The testing bottle method is used to determine the bacterial counts in water using the principle of extinction dilution method.

Testing method:
Identification  of characteristics  of bacterial  growth  display:The  liquid  in the SRB Test Bottle turning black,with black precipitates or fine iron nails turning black all indicate the growth of SRB.
The liquid in the saprophytic bacteria TGB test bottle changes from red to yellow,or the original liquid changes from clear and transparent to turbid and opaque,both indicating the growth of heterotrophic bacteria.
The presence of black or brown colloidal precipitates in the liquid of the iron bacteria IB test bottle,or the disappearance of the original red brown color to a transparent clear liquid,all indicate the growth of iron bacteria.
